Sure, Charles.
Name both the internal and external drives the same ("Macintosh HD" or whatever you're using).
Then, use "Backup - all files" to copy from the internal to the external.
Boot from the external, and make sure everything's the way you want it to be.
Once you're sure, swap the drives, then, use SuperDuper! to do "Backup - all files" back from the external to the internal (once again, named the same).
Finally, shut down, detach the external and start up from the internal.
That should work great!
