Yes, it will. Note that it's best to try to checkpoint any databases before backing up (e.g. mysqldump) and do your copy when the server is as idle as possible (or offline).

I have my 10.4.7 OSX server backing up to 1 internal drive, and one external FireWire drive, all drives boot and run perfectly, and access to them via clients is seemless.
